I have a 2 node ESX cluster running in a lab, and each host had one single physical disk in it when they were created. I've since added a second hard drive into each host and I'd like to add them into the VSAs I already have running. Is it possible to create a VMDK on the second disk and present it to the VSA VM as SCSI 1:1 or do I have to create a whole new VSA just to be able to use these disks?

You have to create new VSA to consume the new disk.
The current VSA only allows 1 VMDK (SCSI 1:0) for data storage.
